Project-description:

The aim of this project is a further characterization of protein pathways altered in neurodegenerative diseases such as Parkinson's disease and Huntington's disease. Using Y2HS we have recently identified proteins that link both pathways. These proteins need now to be investigated if their interaction is altered by the polQ expansion causing HD or by mutations of the PD pathway. One of the interactors is periphilin, which interacts with synphilin 1, an interactor of alpha-synuclein. Using numerous transgenic and knock out models of the PD and HD pathways, we will study whether these interactors are up- or down-regulated in the brain of the mice and rat models. If we identify such differential regulation we will overexpress or downregulate by RNAi these proteins in cell culture and study the behaviour of the cells in respect to aggregation formation and cell death.
